Background | Receptor tyrosine-protein kinase erbB-3, also known as HER3 (human epidermal growth factor receptor 3), is a membrane bound protein that in humans is encoded by the ERBB3 gene.ErbB3 is a member of the epidermal growth factor receptor (EGFR/ERBB) family of receptor tyrosine kinases. The kinase-impaired ErbB3 is known to form active heterodimers with other members of the ErbB family, most notably the ligand binding-impaired ErbB2.ERBB3 (Erb-B2 Receptor Tyrosine Kinase 3) is a Protein Coding gene. Diseases associated with ERBB3 include Lethal Congenital Contracture Syndrome 2 and Erythroleukemia, Familial. Among its related pathways are PI3K/AKT activation and RET signaling. Gene Ontology (GO) annotations related to this gene include protein homodimerization activity and transferase activity, transferring phosphorus-containing groups. An important paralog of this gene is ERBB4. |
Description | A DNA sequence encoding the mouse ERBB3 (Q61526) extracellular domain (Met 1-His 641) was expressed, with a C-terminal His tag. |
Source | HEK293 |
Predicted N Terminal | Ser 20 |
AA Sequence | Met 1-His 641 |
Molecular Weight | The secreted recombinant mouse ERBB3 comprises 633 amino acids and has a calculated molecular mass of 70 kDa. As a result of glycosylation, the apparent molecular mass of the recombinant protein is approximately 95 kDa in SDS-PAGE under reducing conditions. |
Purity | >93% as determined by SDS-PAGE |
Endotoxin | < 1.0 EU per μg of the protein as determined by the LAL method |
Bioactivity | Measured by its binding ability in a functional ELISA.1. Immobilized mouse ErbB3-His at 10 ug/ml (100 ul/well) can bind biotinylated human NRG1 (isoform Beta1) with a linear range of 0.018-0.125 ug/ml.2. Immobilized mouse ErbB3-His at 10 ug/ml (100 ul/well) can bind biotinylated human NRG1 (aa 2-246)-Fc with a linear range of 6.25-100 ng/ml. |
