Background | PIP4K2B (phosphatidylinositol-5-phosphate 4-kinase, type II, beta) catalyzes the phosphorylation of phospha-tidylinositol-5-phosphate on the fourth hydroxyl of the myo-inositol ring to form phosphatidylinositol-5,4-bisphosphate. PIP4K2B is a member of the phosphatidylinositol-5-phosphate 4-kinase family. PIP5K2B interacts specifically with the juxtamembrane region of the p55 TNF receptor and that PIP5K2B activity is increased in mammalian cells by treatment with TNF-alpha (1). PIP5P4KB plays a role in determining insulin sensitivity and adiposity (2). |

Target Details
Target Function | Participates in the biosynthesis of phosphatidylinositol 4,5-bisphosphate. Preferentially utilizes GTP, rather than ATP, for PI(5)P phosphorylation and its activity reflects changes in direct proportion to the physiological GTP concentration. Its GTP-sensing activity is critical for metabolic adaptation. PIP4Ks negatively regulate insulin signaling through a catalytic-independent mechanism. They interact with PIP5Ks and suppress PIP5K-mediated PtdIns(4,5)P2 synthesis and insulin-dependent conversion to PtdIns(3,4,5)P3. |
