Product Overview
Tag | His&Fc |
Host Species | Human |
Accession | NP_036224.1 |
Synonym | ICOS Inducible T-cell costimulator; Activation-inducible lymphocyte immunomediatory molecule; CD28 and CTLA-4-like protein; CCLP; CD28-related protein 1; CRP-1; CD279; Icos; Ailim |
Background | Inducible Costimulator (ICOS) is a member of the growing CD28 family of immune costimulatory receptors. Other family members are CD28, CTLA4 and PD1. ICOS shares approximately 39% amino acid similarity with CD 28 and CTLA4. Mouse and human ICOS share approximately 72% amino acid identity. ICOS is expressed on most CD45RO+ cells. ICOS expression is up-regulated within approximately 24-48 hours of activation on Th primed cells. B7-H2, a member of the B7 family of costimulatory ligands, has been identified as the ICOS ligand. The B7-H2/ ICOS interaction appears to play roles in T cell dependent B cell activation and Th differentiation. In addition, ICOS is more potent in the induction of IL-11 production, acytokine important for suppressive function of T regulatory cells. |
Description | A DNA sequence encoding the extracellular domain of human ICOS (NP_036224.1) (Met 1-Phe 141) was fused to the C-terminal His-tagged Fc region of human IgG1 at the C-terminus. |
Source | HEK293 |
Predicted N Terminal | Glu 21 |
AA Sequence | Met 1-Phe 141 |
Molecular Weight | The recombinant human ICOS/Fc is a disulfide-linked homodimeric protein. The reduced monomer consists of 368 amino acids and predicts a molecular mass of 41.6 kDa. The apparent molecular mass of rhICOS/Fc monomer is approximately 50 kDa due to glycosylation. |
Purity | Greater than 90% as determined by SDS-PAGE |
Endotoxin | < 1.0 EU per μg of the protein as determined by the LAL method |
Bioactivity | Measured by its binding ability in a functional ELISA. Immobilized human human B7-H2 at 1 ug/ml (100 ul/well) can bind human ICOS with a linear range of 1.6-200 ng/ml. |
Formulation | Lyophilized from sterile PBS, pH 7.4 |
Stability | Recombinnat Proteins are stable for up to 1 year from date of receipt at -70°C |
Usage | For Research Use Only |
Storage | Store recombinant protein under sterile conditions at -20°C to -80°C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles. |
