Synonym ERBB4, ALS19, HER4, p180erbB4, erb-b2 receptor tyrosine kinase 4
Background Receptor tyrosine-protein kinase erbB-4 is an enzyme that in humans is encoded by the ERBB4 gene. Alternatively spliced variants that encode different protein isoforms have been described; however, not all variants have been fully characterized. Receptor tyrosine-protein kinase erbB-4 is a receptor tyrosine kinase that is a member of the epidermal growth factor receptor family. ERBB4 is a single-pass type I transmembrane protein with multiple furin-like cysteine rich domains, a tyrosine kinase domain, a phosphotidylinositol-3 kinase binding site and a PDZ domain binding motif. The protein binds to and is activated by neuregulins-2 and -3, heparin-binding EGF-like growth factor and betacellulin. Ligand binding induces a variety of cellular responses including mitogenesis and differentiation. Multiple proteolytic events allow for the release of a cytoplasmic fragment and an extracellular fragment.
Description A DNA sequence encoding the human ERBB4 (Met1-Arg649) was expressed in Baculovirus-Insect Cells, fused with a C-terminal His tag. Human and Rhesus ERBB4 sequences are identical.
Source Insect Cells
Predicted N Terminal Gln 26
AA Sequence Met1-Arg649
Molecular Weight The recombinant human ERBB4 comprises 635 amino acids and has a predicted molecular mass of 71.1 kDa. The apparent molecular mass of the protein is approximately 92 kDa in SDS-PAGE under reducing conditions.
Purity >95% as determined by SDS-PAGE
Endotoxin < 1.0 EU per μg of the protein as determined by the LAL method
Bioactivity Measured by its binding ability in a functional ELISA. Immobilized human ERBB4-His (1-649) at 10 ug/ml (100 ul/well) can bind biotinylated human NRG1, The EC50 of biotinylated human NRG1 is 0.49-1.13 ug/ml.
Formulation Recombinant Human ErbB4 Protein (His Tag) was lyophilized from sterile PBS, pH 7.4.
Stability Recombinant Human ErbB4 protein is stable up to 1 year at -70°C
Usage For Research Use Only
Storage Store the protein under sterile conditions at -20°C to -80°C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.
