| Description | Native Human Antithrombin III functions as the principal serine protease inhibitor of the coagulation cascade, exerting its anticoagulant activity through a dual mechanism involving direct, progressive inhibition of activated clotting factors—most notably thrombin (Factor IIa) and Factor Xa—and a dramatic, template-enhanced inhibition when bound to heparin or heparan sulfate proteoglycans on vascular endothelial surfaces. This critical regulator maintains blood fluidity by forming stable 1:1 covalent complexes with target proteases, a process accelerated over 1000-fold by heparin cofactor II-mediated conformational changes that expose its reactive center loop, thereby preventing excessive fibrin formation and thrombus propagation. Through its sophisticated control of thrombin generation and activity, Antithrombin III not only serves as a natural defense against thrombosis but also modulates inflammatory responses through protease inhibition, with its hereditary or acquired deficiency directly predisposing to venous thromboembolism and its purified form constituting a cornerstone therapeutic for heparin resistance and acute thrombotic events. |
