| Description | Native Human Alpha-2-HS-Glycoprotein (AHSC; Fetuin-A) functions as a multifunctional plasma glycoprotein that plays pivotal roles in mineral metabolism, bone remodeling, and the regulation of insulin signaling through its dual capacity to inhibit ectopic calcification and modulate cytokine activity. This hepatocyte-derived protein, which belongs to the cystatin superfamily, operates as a systemic inhibitor of pathological calcification by forming soluble complexes with calcium and phosphate ions to prevent precipitation, while simultaneously serving as a natural antagonist of TGF-β signaling through direct binding to the cytokine and its receptors. Through these mechanisms, fetuin-A not only maintains mineral homeostasis and vascular health but also influences insulin receptor tyrosine kinase activity, inflammatory responses, and macrophage activation, with its plasma concentrations serving as a biomarker for insulin resistance, cardiovascular calcification risk, and overall mortality in chronic kidney disease and systemic inflammatory conditions. |
