| Description | Native Human Alpha-2-Antiplasmin serves as the primary and most rapid physiological inhibitor of fibrinolysis, functioning through a dual mechanism that immediately neutralizes free plasmin in circulation while simultaneously being incorporated into fibrin clots to confer specific protection against premature dissolution. This serpin-family glycoprotein employs a unique reactive center that specifically targets the active site of plasmin, forming a stable 1:1 covalent complex that irreversibly inactivates the protease, while its cross-linking to fibrin by Factor XIIIa during coagulation creates fibrinolysis-resistant clots through competitive inhibition of plasminogen binding. Through this sophisticated regulation of plasmin activity, alpha-2-antiplasmin maintains the crucial balance between coagulation and fibrinolysis, with its congenital deficiency resulting in a severe hemorrhagic diathesis characterized by uncontrolled clot breakdown, while its consumption during hyperfibrinolysis serves as an important clinical marker for conditions such as disseminated intravascular coagulation and thrombolytic therapy monitoring. |
