| Background | Carboxylesterase 2 (CES2) is a member of the carboxylesterase family, which catalyzes the hydrolysis transesterification of various xenobiotic and endogenous substrates with ester, thioester, or amide bonds to detoxify the xenobiotics and activate the prodrugs (1). CES2 is mainly expression in intestine to hydrolyze substrates with a large alcohol group and small acyl group such as aspirin, the anticancer prodrug irinotecan (CPT-11), and flutamide (2). In addition, CES2 is also involved in obesity and fatty liver disease, which is related in lipid metabolism (2). |

| Target Function | Involved in the detoxification of xenobiotics and in the activation of ester and amide prodrugs. Shows high catalytic efficiency for hydrolysis of cocaine, 4-methylumbelliferyl acetate, heroin and 6-monoacetylmorphine. Hydrolyzes aspirin, substrates with large alcohol group and small acyl group and endogenous lipids such as triacylglycerol. Converts monoacylglycerides to free fatty acids and glycerol. Hydrolyzes of 2-arachidonoylglycerol and prostaglandins. |
