Human papillomavirus (HPV) L1 is the major capsid protein that self-assembles into virus-like particles (VLPs), which are the basis for all current prophylactic HPV vaccines. HPV L1 Target Overview

L1 is a 55-60 kDa protein that forms pentameric capsomeres; 72 pentamers assemble into a T=7 icosahedral VLP. L1 is highly immunogenic and contains type-specific conformational epitopes that are the targets of neutralizing antibodies. HPV L1 plays key roles in:

  • Viral attachment and entry via heparan sulfate proteoglycans (HSPG) on host cells

Over 200 HPV types exist, with types 16, 18, 31, 33, 45, 52, 58 classified as high-risk for cervical cancer. L1 sequence diversity is high, making type-specific selection critical for vaccine and serological studies.
