Synonym ERBB2, CD340, HER-2, HER-2/neu, HER2, MLN 19, NEU, NGL, TKR1, erb-b2 receptor tyrosine kinase 2
Background Receptor tyrosine-protein kinase erbB-2, also known as CD340 (cluster of differentiation 340), proto-oncogene Neu, Erbb2 (rodent), or ERBB2 (human), is a protein that in humans is encoded by the ERBB2 gene. ERBB is abbreviated from erythroblastic oncogene B, a gene isolated from avian genome. It is also frequently called HER2 (from human epidermal growth factor receptor 2) or HER2/neu. ERBB2, commonly referred to as HER2, is amplified and/or overexpressed in 20-30% of invasive breast carcinomas. HER2-positive breast cancer is treated in a separate manner from other subtypes of breast cancer and commonly presents as more aggressive disease. Metastatic HER2-positive breast cancer is now commonly treated with HER2-targeted therapy. Apart from being amplified/overexpressed, ERBB2 activating mutations have been shown to have clinical importance in HER2-negative breast cancer. These mutations have shown sensitivity to the tyrosine kinase inhibitor neratinib, and highlight the importance of clinical sequencing efforts in treating breast cancer.
Description A DNA sequence encoding the canine ERBB2 (Met1-Thr652) was expressed with a C-terminal His tag.
Source HEK293
Predicted N Terminal Thr 23
AA Sequence Met1-Thr652
Molecular Weight The recombinant canine ERBB2 comprises 641 amino acids and has a predicted molecular mass of 70.4 kDa. The apparent molecular mass of the protein is approximately 75- 95 kDa in SDS-PAGE under reducing conditions.
Purity >95% as determined by SDS-PAGE
Endotoxin < 1.0 EU per μg of the protein as determined by the LAL method
Bioactivity 1.Measured by its binding ability in a functional ELISA. 2.Immobilized canine ErbB2 at 10ug/mL (100uL/well) can bind herceptin, the EC50 of herceptin is 10-50ng/mL.
Formulation Recombinant Canine Her2 / ERBB2 Protein (His Tag) was lyophilized from sterile PBS, pH 7.4.
Stability Recombinant Canine Her2 / ERBB2 protein is stable up to 1 year at -70°C
Usage For Research Use Only
Storage Store the protein under sterile conditions at -20°C to -80°C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.
